VMFS6 added support for 4K native (4Kn) drives, which use a 4096-byte physical sector size; VMFS5 only supported 512-byte and 512e drives and could not address 4Kn devices at all.
512e (512-byte emulated) disk support was already present in VMFS5, so it is not a new VMFS6 enhancement.
GPT (GUID Partition Table) partitioning for storage devices was already used by VMFS5 for datastores on devices larger than 2 TB, making it not exclusive to VMFS6.
